Structure Therapeutics Inc. on Tuesday announced encouraging clinical trial results for two of its lead oral candidates targeting metabolic diseases and chronic weight management.

Topline results from the early-stage trial of ACCG-2671 and 72-week extension study data for aleniglipron showed significant weight reduction and favorable tolerability.

Early Data for ACCG-2671 Positive

The Phase 1/2a single ascending dose study of ACCG-2671 evaluated 31 healthy, non-obese participants, with doses ranging from 1 mg to 10 mg.

The candidate demonstrated rapid absorption and a terminal half-life of approximately six days, supporting daily or weekly dosing.

Participants receiving a single 10 mg dose achieved a mean weight reduction of 3.3% by Day 24.

Additionally, the treatment reduced the bone resorption biomarker CTX-1 by approximately 60% at Day 2.

No serious adverse events or liver enzyme elevations were recorded in the trial, though dose-related gastrointestinal side effects were observed at higher doses.

Based on these results, the company initiated a multiple ascending dose trial, with data expected in the first half of 2027.

Aleniglipron Achieves Sustained Weight Loss

In the ACCESS open-label extension study, aleniglipron demonstrated sustained weight reduction through 72 weeks in adults continuing daily oral treatment.

Patients receiving the maximum 180 mg dose achieved weight reductions of up to 16.2%, without reaching a plateau.

Initiating treatment at a lower 2.5 mg dose enhanced gastrointestinal tolerability, and fewer than 5% of participants discontinued treatment due to adverse events.

Ongoing Milestones and Phase 3 Program

These findings support the ongoing Phase 3 ACCOMPLISH program evaluating aleniglipron, which launched in August 2026, with topline data expected in the second half of 2028.

Additionally, the company expects to report results from three distinct trials in the fourth quarter of 2026, focusing on body composition, type 2 diabetes, and patients transitioning from injectable GLP-1 medications to oral aleniglipron.
