Addressing Unmet Needs in Oncology
AKTX-101 is a TROP2-targeted ADC that utilizes a proprietary PH1 RNA spliceosome-modulating payload.
According to the company, this novel mechanism addresses emerging shortcomings of existing ADC payload classes.
Urothelial carcinoma represents a rapidly expanding market for ADC therapies, underscored by the success of treatments such as Padcev.
Despite these advances, patients who relapse after standard therapy face a severe lack of viable treatment options.
Encouraging Preclinical Tumor Suppression
In recent evaluations, AKTX-101 demonstrated meaningful antitumor efficacy across various clinically relevant models.
In the UM-UC-14 advanced urothelial carcinoma model—representing the metastatic setting where Padcev serves as a first-line standard-of-care option—AKTX-101 achieved statistically significant tumor growth inhibition, while Padcev showed limited effect.
Additionally, the experimental therapy showed encouraging results against tumors that had developed resistance to Trodelvy. When these resistant tumors were treated with AKTX-101, growth was markedly slowed.
This suggests that the PH1 payload's ability to disrupt RNA splicing can overcome resistance associated with traditional topoisomerase I inhibitors, potentially offering benefits in lung and breast cancers.
Future Clinical Milestones
Akari continues to advance the pre-IND process for AKTX-101. The oncology biotech company plans to initiate a Phase 1 clinical trial in mid-2027, while also exploring further development opportunities for its PH1 payload platform to maximize its commercial potential.
