Enliven Therapeutics Inc. received an Outperform rating from William Blair on Tuesday, which initiated coverage, citing the potential of its lead drug candidate ELVN-001 to reshape the treatment landscape for chronic myeloid leukemia.

ELVN-001 Shows Positive Phase 1 Data

Enliven Therapeutics' lead drug, ELVN-001, is a small-molecule kinase inhibitor designed to specifically target the BCR::ABL1 gene fusion, the oncogenic driver of CML.

In June, the company reported positive data from its ongoing Phase 1 ENABLE clinical trial, which evaluates ELVN-001 in previously treated CML patients.

The data showed an overall major molecular response of 61% in the 80 mg once-daily Phase 1b cohort, with a 48% MMR rate at 24 weeks.

Response rates were higher in earlier-line patients, with an overall MMR of 67% in patients with only one or two prior distinct tyrosine kinase inhibitors, and 55% achieving MMR at 24 weeks.

Enliven Therapeutics Cash Reserves to Sustain Through 2030

As of June 30, 2026, the company held cash, cash equivalents, and marketable securities totaling $895.2 million, which is expected to provide a cash runway through 2030.

William Blair noted that ELVN-001 has the potential to serve patients across the treatment continuum, with an initial focus on the growing late-line patient population, followed by expansion into earlier treatment settings.

William Blair Bullish on $8.8 Billion CML Market

Analyst Andy Hsieh wrote that despite multiple approved CML tyrosine kinase inhibitors on the market, unmet medical needs remain, as a growing number of late-line patients seek more effective, better-tolerated, and more convenient treatment options.

William Blair estimates the current total addressable market for CML at approximately $8.8 billion, with the U.S. market at $5.6 billion.

The analyst also noted that at the planned Phase 3 dose level, the 48% MMR achievement rate is twice the rate demonstrated by Novartis AG's Scemblix, the current market leader.

Additionally, ELVN-001 is the only TKI shown to have activity against newly emerging resistance mutations to Scemblix treatment.

The analyst stated that ELVN-001 could disrupt the chronic myeloid leukemia treatment landscape, citing its differentiated profile and potential to serve patients across multiple treatment lines.

ELVN stock performance: As of Tuesday's data release, Enliven shares were down 3.80% at $1.52.
