On Tuesday, Pharvaris NV announced positive top-line results from a late-stage clinical trial evaluating its oral drug deucrictibant extended-release tablets for the prevention of hereditary angioedema attacks.

Following the announcement, the company's shares surged more than 17%.

The trial met all primary and secondary efficacy endpoints, paving the way for a regulatory submission planned for the first half of 2027.

Rapid Onset and Durable Protection

This global, double-blind CHAPTER-3 trial evaluated a daily 40 mg dose of deucrictibant XR versus placebo over a 24-week period.

The study enrolled 85 participants. Notably, it is the only Phase 3 prevention study to include patients with type 1 HAE, type 2 HAE, and those with normal C1 inhibitor levels.

Compared to the placebo group, patients receiving the daily tablet experienced a 83% reduction in the mean monthly attack rate.

For the 80 participants specifically diagnosed with type 1 or type 2 HAE, the therapy reduced the monthly attack rate by 87%.

Clinical data indicated that participants achieved protection within the first week of treatment. This protective effect was sustained throughout the entire 24-week study period, leading to a substantial increase in the number of attack-free patients.

Favorable Safety Profile and Next Steps

The treatment was well-tolerated by participants. Researchers observed mostly mild to moderate adverse events, with no treatment-related serious adverse events reported.

Only one patient in each study group discontinued the trial due to an adverse event.

Following these successful results, the company plans to submit a new drug application to the U.S. Food and Drug Administration in early 2027. Meanwhile, the CHAPTER-4 open-label extension study is ongoing to monitor long-term effects.

Additionally, the company expects to report top-line data from a separate Phase 3 trial for acquired angioedema in the first quarter of 2027.
